In present day, a genius physicist named Sento Kiryū is hired by the government-funded Touto Institute of Advanced Matter Physics in uncovering the secrets of the Pandora Box. But Sento, having no memory of his past other than being held captive by the mysterious Night Rogue of the Faust organization, secretly moonlights on the side as Kamen Rider Build to fight the Smash monsters that are created by Faust's experimentations.

History 10 years ago, the “Pandora Box” was found on Mars. On Earth, it created walls that divided Japan into Touto, Hokuto and Seito and brought along the Smash, unidentified lifeforms that attack people. Sento Kiryu, prodigy physicist, transforms into Kamen Rider Build to fight the Smash. The Touto government requests his help in deciphering the mystery behind the Pandora Box and the Smash. There is one mystery not even Sento can solve his own past.

Pandora Box Is an extraterrestrial artifact of unknown origin that was found during a manned Mars mission. The alien artifact was found by Soichi Isurugi, who was a crew member of the INSET exploration vessel Prominence during the astronaut's survey of Martian caverns at the time. Upon being touched, it reacted to the human by shedding its stone encasing to reveal its true form. The box was then carried back onto the Prominence and back to Earth. The Pandora Box was believed to have been technology from part of an advanced civilization on Mars.

Prime Ministers As a result of the Skywall Disaster in 2007, Japan has been divided into three different districts, namely: Touto, Hokuto, Seito. Everyone present at the event was suspected by Himuro to become much more aggressive. As a result of the separation, several foreign powers have attempted to take advantage of the situation although neither of the districts display interest in allying with external forces.

Nascita Nascita is a café owned by Sōichi Isurugi where he lives with his daughter Misora. After meeting Sento, they shelter him there as well as allow him to live in the basement. They also shelter Ryūga after Sento rescues him from the authorities. However, the revelation of Sōichi Isurugi being Blood Stalk compromises the café's status as a safehouse for a short while until he leaves Touto.
  • Soichi Isurugi / Blood Stalk : Is the rather enigmatic 48-year old owner of nascita and Misora's father. Ten years prior to the series, he was the astronaut who discovered the Pandora Box on Mars and caused the Skywall Disaster by coming in contact with the artifact.
  • Sento Kiryu / Kamen Rider Build : Physics scientist and Pandora Box researcher.
  • Ryuga Banjyou / Kamen Rider Cross-Z : Is an ex-fighter who was disqualified for cheating. He was convicted of the murder of a scientist involved with the Pandora Box and broke out of jail.
  • Misora Isurugi : Sento's lab assistant and daughter of Soichi Isurugi
  • Sawa Takigawa : Is a journalist investigating the Pandora Box. She aids Sento Kiryu by providing him with information.
Touto Institute of Advanced Matter Physics Is an organization appearing in Kamen Rider Build. They are an advanced scientific government research laboratory with some of its staff being officials in Touto's government who deliver reports directly to the Prime Minister of Touto.
  • Gentoku Himuro / Night Rogue : Is the 35-year old Special Advisor to his father Prime Minister Taizan Himuro and head of the Touto Institute. While having been exposed to the Pandora Box's radiation.
  • Nariaki Utsumi / Fake Night Rogue : Is the 28-year old secretary of Himuro who was once a scientist of the Touto Institute. Little is known about him and his personality other than the fact that he is highly loyal to Gentoku and is a scientist of Faust.
  • Sento Kiryu / Kamen Rider Build : Physics scientist and Pandora Box researcher.
  • Takumi Katsuragi : Was a researcher at Touto Institute of Advanced Matter Physics who was murdered by Soichi Isurugi upon his intended defection from Faust, an organization he founded.
  • Shingo Kuwata : Is a researcher in the Touto Institute who is one of Katsuragi's sympathizers. He uses his Fullbottle to become his own version of the Flying Smash.
  • Eita Kawai : Is a researcher in the Touto Institute who is one of Katsuragi's sympathizers.
  • Kaisei Mogami : Is a cyborg, Mogami was a researcher in the Touto Institute of Advanced Matter Physics and an associate of Takumi Katsuragi.
Hokuto Troopers
  • Kazumi Sawatari / Kamen Rider Grease : Is a 29-year old soldier from Hokuto who was originally a farm owner until the Skywall Disaster ruined his livlihood as he sacrificed his life savings to help out his Hokuto Three Crows.
Hokuto Three Crows Are a group of Hokuto soldiers who were originally workers of Kazumi Sawatari's farm and enlisted into Hokuto's military to repay their former employer's debt for financially supporting their families. Each are able to transform into enhanced form of the Smash known as Hard Smash and later the stronger Hazard Smash.
  • Akaba / Masaru Ooyama / Castle Hard Smash : That has the Catapulta Cannon on his head and the two Ground Rampart shields on his shoulder pads.
  • Aoba / Syuuya Aikawa / Stag Hard Smash : That is armed with two sword-like Rapture Scissors. Aoba is killed during the battle with Build and his newly acquired RabbitTank Hazard form
  • Kiba / Syoukichi Mihara / Owl Hard Smash : That has the two Forest Seeker drones on his gauntlets. He has a childish behavior, even acting literally like a child throwing a fit when defeated by Build in one battle.
Nanba Heavy Industries Nanba Heavy Industries is a company which provides financial support to Faust.
  • Jūsaburō Nanba : Is the corrupted and war-mongering chairman of Nanba Heavy Industries. Nanba's only reason of collaborating with Faust is to acquire the Build system and use it for military applications.
  • Nariaki Utsumi / Fake Night Rogue : Is the 28-year old secretary of Himuro who was once a scientist of the Touto Institute. Little is known about him and his personality other than the fact that he is highly loyal to Gentoku and is a scientist of Faust.
Faust Is a mysterious terrorist organization who conduct experiments on humans to create the Smash creatures. It was originally founded by Touto's former scientist Takumi Katsuragi and Gentoku Himuro. They are in possession of one of the Pandora Box's panels that was split during the Skywall Disaster, along with Nebula Gas they extracted from the Skywall and liquefied into the element used in the development of Fullbottles.

Guardians Are enforcement androids mass-produced by Nanba Industries as peacekeepers of each countries. At first, only Touto is seemingly able to build Guardians, until the revelation that Nanba has been supporting the other two Governments as well. They also act as sleeper agents of Faust and their true programming can be activated by Blood Stalk.

Smash Are monsters who were originally humans that have been injected with the liquefied Nebula Gas as part of Faust's experiments. This process results in the experimented losing almost every remaining memories of the victims have left. In order to revert a Smash back to a normal human, the Kamen Riders must defeat them and extract their element liquids into an Empty Bottle which can then be purified into a Fullbottle for their transformation and equipment uses.
